Pseudomonas putida as universal biocatalyst for the selective oxidation of monoterpenes

The solvent-tolerant bacterium Pseudomonas putida will be optimized by metabolic engineering to meet the challenges of whole-cell biotransformations of hydrophobic and toxic substances such as low biocatalyst activity and stability. The monoterpenes α-pinene and limonene serve as model substances.

The resulting whole-cell biocatalyst represents a platform organism for the expression of cofactor-dependent oxygenases and subsequent use in biotransformations in aqueous-organic two-phase systems. Oxidation of organic compounds is of special interest for the chemical and pharmaceutical industry and this system allows the regio- and stereoselective oxidation beyond the family of monoterpenes. Direct biotechnological access to terpenoids can be established for small- and medium-sized enterprises of the pharmaceutical and fine chemistry industry.
