Cofactor Substitution: Mediated Electron Transfer to Enzymes.

A. O. Magnusson & D. Holtmann

Enzymes have become valuable tools in asymmetric synthesis due to their high versatility and specificity as well as the mild reaction conditions required, specially benefitting synthesis with sensitive or reactive compounds. The use of oxidoreductases is steadily increasing due to their interesting reduction, oxidation, and oxyfunctionalization reactions. A “dilemma” utilizing oxidoreductases is their need for redox equivalents. In nature are these redox equivalents supplied through cofactors, generally NAD(P)H to deliver electrons and NAD(P)+ to accept electrons, which are regenerated through the cell metabolism consuming an energy source such as glucose.
